Indirect Somatic Embryogenesis: An Efficient and Genetically Reliable Clonal Propagation System for Ananas comosus L. Merr. Hybrid “MD2”

چکیده
The objective of this study was to establish an efficient—direct or indirect—regeneration system for pineapple (Ananas comosus L.) plants, with a high rate multiplication and that would preserve the genetic identity donor genotype (Hybrid ‘MD2’) in regenerated plants. Ten treatments, different concentrations 2,4-Dichlorophenoxyacetic (2,4-D) Picloram (P), absence presence 6-Benzylaminopurine (BAP), were used vitro morphogenesis induction, as well histological molecular techniques, order characterize morphogenic responses induced. Significant differences between treatments tested, induce callus buds, assessed by Kruskal Wallis method Mann–Whitney U-tests. Different regeneration routes identified, showing potential species. medium containing 2 mg L−1 2,4-D BAP, where indirect somatic embryogenesis occurred, selected most efficient treatment, average 120 embryos per explant, differing significantly from rest treatments. It also demonstrated plants preserved genotype, which represents degree confidence application A. comusus clonal propagation.

Chlorophyll and Protein Interrelationships in Ananas comosus (L.) Merr.
Studies on a quantitative separation and analysis of chloroplastic matter by different investigators have revealed that chlorophyll is held by chemical forces on proteinaceous matter. This matter, named phyllochlorin by MESTRE (17), chloroplastin by STOLL (37), photosynthin by FRENCH (8), and chloroglobin by RABINOWITCH (21), is presumably identical with the substance composing the stroma of ch...
متن کاملEffects of Different Amounts of Potassium on Growth and Ash Constituents of Ananas Comosus (l.) Merr.
The data reported in this paper were obtained from plants grown in nutrient solutions supplied with high or low amounts of potassium, namely 205 or 4 mg. per liter but with equal amounts of either nitrateor ammonium-nitrogen. Tops or crowns (vegetative organs produced at the apex of fruits) of uniform size from a single clone were placed in tap water for one month until they had produced about ...
متن کاملMicropropagation and Growth of in Vitro Pineapple (ananas Comosus L. Merr) in Iran
Although, several pineapple micropropagation protocols have already been published, significant improvement could be achieved, if the stages of in vitro culture were better defined. Our work concerned several experiments aiming at the mass production of high quality plantlets. Tissue culture experiments were therefore conducted to develop rapid multiplication procedures for Ananas comosus L. (M...
متن کاملCloning and Expression Analysis of a PISTILLATA Homologous Gene from Pineapple (Ananas comosus L. Merr)
PISTILLATA (PI)-like genes are crucial regulators of flowering in angiosperms. A homologue of PI, designated as AcPI (Genbank accession number HQ717796), was isolated from pineapple cultivar Comte de Paris by reverse transcriptase polymerase chain reaction (RT-PCR) and rapid amplification of cDNA ends (RACE). The cDNA sequence of AcPI is 907 bp in length and contains an open reading frame of 59...
متن کاملCharacterization of Phosphoenolpyruvate Carboxykinase from Pineapple Leaves Ananas comosus (L.) Merr.
Phosphoenolpyruvate carboxykinase has been partially purified from pineapple (Ananas comosus [L.]) leaves. Specific activities obtained show it to be a major activity in this tissue. Above 15 C, the respective activation energies for decarboxylation and carboxylation are 13 and 12 kcal/mol. Below 15 C, there are discontinuities in Arrhenius plots with an associated large increase in activation ...
